چکیده

Background Talimogene laherparepvec (T-VEC) is an injectable modified oncolytic herpes simplex virus type-1 (HSV-1) hypothesized to be efficacious by at least two complimentary mechanisms of action: a) direct oncolysis of the injected tumor and b) elicitation of a systemic antitumor immune response against non-injected lesions and metastases. The purpose of this study was to test that hypothesis that direct viral oncolysis was not involved in the regression of contralateral (non-injected) tumors.

Local Delivery of OncoVEX Generates SystemicAntitumor ImmuneResponsesEnhanced by Cytotoxic T-Lymphocyte–Associated Protein Blockade

Purpose: Talimogene laherparepvec, a new oncolytic immunotherapy, has been recently approved for the treatment of melanoma. Using a murine version of the virus, we characterized local and systemic antitumor immune responses driving efficacy in murine syngeneic models. Experimental Design: The activity of talimogene laherparepvec was characterized against melanoma cell lines using an in vitro vi...

Talilmogene Iaherparepvec generates systemic T-cell-mediated anti-tumor immunity

Talimogene laherparepvec is an investigational oncolytic immunotherapy based on a modified herpes simplex virus type-1 (HSV-1) that is designed to selectively replicate in tumor tissue and to stimulate a systemic antitumor immune response. Intralesional administration of talimogene laherparepvec is intended to result in oncolysis within injected tumors. Iterative viral replication of virus with...

Tumor regression induced by intratumor therapy with a disabled infectious single cycle (DISC) herpes simplex virus (HSV) vector, DISC/HSV/murine granulocyte-macrophage colony-stimulating factor, correlates with antigen-specific adaptive immunity.

Direct intratumor injection of a disabled infectious single cycle HSV-2 virus encoding the murine GM-CSF gene (DISC/mGM-CSF) into established murine colon carcinoma CT26 tumors induced a significant delay in tumor growth and complete tumor regression in up to 70% of animals. Pre-existing immunity to HSV did not reduce the therapeutic efficacy of DISC/mGM-CSF, and, when administered in combinati...

Reovirus oncolysis: a brief insight on molecular mechanism and immunological aspect

Abstract :  Reovirus (respiratory enteric orphan virus), a naturally occurring benign human pathogen, has an inherent ability to target transformed and cancerous cells and cause their lysis, while leaving non-transformed cells relatively unaffected.  The efficiency of this innate oncolytic activity of reovirus correlates with expression of the ras oncogene.  Cells expressing ac...

Encoding CD40 Ligand Intratumoral Injection of Recombinant Fowlpox Virus Therapeutic Vaccination against Murine Lymphoma by Updated Version

The interaction between CD40 ligand (CD40L, CD154) and its receptor CD40 on antigen-presenting cells is essential for the initiation of cell-mediated and humoral immune responses. Malignant B cells also express CD40 and respond to CD40L by enhancing expression of costimulatory molecules. In this study, we investigated the therapeutic antitumor effect of intratumoral administration of recombinan...
